                        <title>Synthesis, Characterization, and Anticancer Activity of Heterocyclic and Coordination Compounds</title>

                        <abstract language="eng"><p>Heterocyclic compounds and metal coordination complexes constitute two important classes of molecules in contemporary medicinal chemistry because of their structural diversity, tunable physicochemical properties, and broad spectrum of biological activities. The incorporation of heteroatom-containing rings into bioactive molecular frameworks can enhance molecular recognition, solubility, lipophilicity, and interaction with biological targets, whereas coordination with metal ions can introduce additional mechanisms of biological action, including redox modulation, DNA interaction, enzyme inhibition, mitochondrial dysfunction, and generation of reactive oxygen species. This review discusses recent developments in the synthesis, characterization, and anticancer evaluation of heterocyclic compounds and their metal coordination complexes. Conventional and modern synthetic approaches, including cyclization, condensation, Schiff-base formation, multicomponent reactions, and ligand-assisted metal coordination, are examined. Characterization using Fourier-transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, ultraviolet-visible spectroscopy, mass spectrometry, elemental analysis, thermal analysis, and single-crystal X-ray diffraction provides essential information regarding molecular structure and coordination geometry. Particular attention is given to transition-metal complexes containing ruthenium, platinum, copper, cobalt, nickel, zinc, and palladium, as well as biologically relevant heterocyclic scaffolds such as imidazole, thiazole, triazole, pyridine, quinoline, indole, benzimidazole, and quinazoline. Their anticancer potential is discussed in relation to cytotoxicity, apoptosis induction, cell-cycle arrest, oxidative stress, DNA binding, and inhibition of cancer-associated enzymes and signaling pathways. The review also highlights challenges associated with selectivity, toxicity, solubility, pharmacokinetics, and clinical translation. Rational ligand design, combination strategies, nanotechnology-assisted delivery, and mechanism-based screening may facilitate the development of next-generation heterocyclic and coordination compounds with improved anticancer efficacy and selectivity.</p>

<p class="wp-block-paragraph"><strong>Introduction</strong></p>



<p class="wp-block-paragraph">Cancer remains one of the major causes of morbidity and mortality worldwide, creating a continuing need for new therapeutic molecules with improved efficacy, selectivity, and safety. Although substantial progress has been achieved through targeted therapies, immunotherapy, and molecularly directed treatments, limitations such as drug resistance, systemic toxicity, tumor heterogeneity, and treatment failure continue to restrict therapeutic outcomes. Consequently, the discovery of structurally diverse small molecules capable of interacting with multiple biological targets remains an important area of pharmaceutical and medicinal chemistry. Heterocyclic compounds represent one of the largest and most important structural classes in drug discovery [1]. Nitrogen-, oxygen-, and sulfur-containing heterocycles occur in numerous clinically important drugs and natural products. Their electronic properties, hydrogen-bonding capacity, aromaticity, and ability to participate in hydrophobic and electrostatic interactions allow them to bind efficiently to biological macromolecules. Coordination chemistry provides another strategy for developing biologically active compounds. Complexation of organic ligands with metal ions can substantially modify the biological properties of the parent ligand. Metal coordination may alter charge, geometry, lipophilicity, redox behavior, stability, and cellular uptake. Importantly, metal complexes may display biological mechanisms that are not accessible to conventional organic molecules. The clinical success of platinum-based drugs demonstrated the potential of metal complexes in oncology. However, adverse effects, acquired resistance, and limited tumor selectivity associated with classical platinum drugs have stimulated extensive research into alternative metals and ligand systems. Ruthenium, copper, cobalt, palladium, zinc, nickel, and other metals have therefore been investigated as potential components of anticancer coordination compounds. The combination of heterocyclic ligands and metal coordination is particularly promising because the heterocyclic framework can provide strong and selective metal-binding sites while simultaneously contributing to biological activity [2]. This review examines the synthesis, structural characterization, and anticancer activity of heterocyclic and coordination compounds, with emphasis on the relationship between molecular structure and biological function.</p>



<p class="wp-block-paragraph"><strong>2. Heterocyclic Scaffolds in Anticancer Drug Discovery</strong></p>



<p class="wp-block-paragraph">Heterocyclic chemistry provides a highly versatile platform for the development of anticancer agents because heteroatoms can modify electronic distribution, molecular polarity, hydrogen-bonding properties, and target recognition. Nitrogen-containing heterocycles are particularly prominent in medicinal chemistry, although oxygen- and sulfur-containing systems are also widely investigated. Pyridine and pyrimidine derivatives can interact with nucleic acids, kinases, and other enzymes and have therefore been extensively incorporated into anticancer drug candidates. Imidazole and benzimidazole scaffolds can participate in hydrogen bonding and π–π interactions and have been investigated for kinase inhibition, DNA interaction, and enzyme modulation. Indole derivatives are structurally important because the indole ring occurs in several biologically active natural products and pharmaceutical molecules. Substitution at different positions can substantially modify their cytotoxic and target-specific properties. Quinoline and quinazoline derivatives have received considerable attention because their planar aromatic structures can facilitate interactions with DNA and ATP-binding sites of protein kinases. Quinazoline-based structures are particularly important in the development of kinase-targeted anticancer agents [3]. Sulfur-containing heterocycles, including thiazoles, thiophenes, and thiosemicarbazone-derived systems, also exhibit considerable biological diversity. Their sulfur atoms can influence metal coordination and redox behavior, making them useful for developing both organic and metal-based anticancer agents.</p>



<p class="wp-block-paragraph"><strong>3. Synthesis of Heterocyclic Compounds</strong></p>



<p class="wp-block-paragraph">The synthesis of heterocyclic compounds involves numerous strategies, depending on the required ring system and substitution pattern. Cyclization reactions remain among the most important approaches and can involve intramolecular condensation, nucleophilic substitution, electrophilic reactions, or transition-metal-catalyzed processes. Schiff-base formation is another widely used strategy, particularly when heterocyclic compounds are intended to function as ligands for metal coordination. Aldehydes or ketones react with primary amines to generate azomethine-containing structures, which can subsequently coordinate with transition-metal ions. Multicomponent reactions have become increasingly important because they allow the construction of structurally complex heterocycles in fewer synthetic steps. These approaches can reduce reaction time, solvent consumption, and purification requirements while providing substantial structural diversity [4]. Other approaches include microwave-assisted synthesis, solvent-free reactions, green synthesis, photocatalytic reactions, and metal-catalyzed cross-coupling. Modern synthetic methodologies increasingly emphasize improved atom economy, reduced waste generation, and environmentally acceptable reaction conditions.</p>



<p class="wp-block-paragraph"><strong>4. Coordination Chemistry of Bioactive Ligands</strong></p>



<p class="wp-block-paragraph">Coordination complexes are formed when a metal center interacts with one or more donor atoms of an organic or inorganic ligand. Common donor atoms include <strong>nitrogen, oxygen, sulfur, and phosphorus</strong>. The coordination environment determines important properties such as geometry, stability, electronic configuration, redox potential, and biological activity. Ligands containing multiple donor sites can form chelate complexes, which are often more stable than complexes involving monodentate ligands. Schiff bases, hydrazones, thiosemicarbazones, pyridines, imidazoles, and quinoline derivatives are frequently used as chelating ligands [5]. Metal coordination can alter the biological behavior of a ligand by modifying its lipophilicity and charge and facilitating interaction with biomolecules. In some systems, complexation can enhance cellular uptake and improve anticancer activity relative to the uncoordinated ligand.</p>



<p class="wp-block-paragraph"><strong>5. Synthesis of Metal Coordination Compounds</strong></p>



<p class="wp-block-paragraph">Metal complexes are commonly synthesized by reacting a preformed ligand with an appropriate metal salt under controlled conditions. The choice of solvent, temperature, pH, reaction time, ligand-to-metal ratio, and counterion can affect the final coordination structure. A general synthetic procedure involves dissolving the ligand in an appropriate solvent followed by addition of the metal salt under continuous stirring. The reaction mixture may subsequently be heated under reflux or maintained at room temperature depending on the stability and reactivity of the reactants. The resulting complex is isolated through filtration, precipitation, evaporation, or crystallization. Transition metals such as ruthenium, platinum, copper, cobalt, nickel, palladium, zinc, and iron have been investigated extensively. Different metal oxidation states and coordination geometries can produce substantially different biological properties [6]. Careful control of stoichiometry is particularly important because the same ligand and metal can generate complexes with different metal-to-ligand ratios and coordination geometries.</p>



<p class="wp-block-paragraph"><strong>6. Characterization of Heterocyclic and Coordination Compounds</strong></p>



<p class="wp-block-paragraph">Comprehensive characterization is essential for confirming the chemical structure, purity, coordination mode, and physicochemical properties of newly synthesized compounds.</p>



<p class="wp-block-paragraph"><strong>6.1 Fourier-transform infrared spectroscopy</strong></p>



<p class="wp-block-paragraph">FTIR spectroscopy provides information regarding functional groups and changes occurring after metal coordination. Shifts in bands associated with <strong>C=N, C=O, N–H, O–H, and C–S</strong> groups may indicate participation of donor atoms in coordination.</p>



<p class="wp-block-paragraph"><strong>6.2 Nuclear magnetic resonance spectroscopy</strong></p>



<p class="wp-block-paragraph">¹H and ¹³C NMR spectroscopy provide information about the chemical environment of atoms within organic ligands. Changes in chemical shifts following metal coordination can provide evidence of ligand binding, although paramagnetic metal ions may complicate NMR interpretation.</p>



<p class="wp-block-paragraph"><strong>6.3 UV-visible spectroscopy</strong></p>



<p class="wp-block-paragraph">UV-visible spectroscopy can provide information about electronic transitions within ligands and metal complexes. Charge-transfer bands and, in some complexes, d–d transitions can provide useful information regarding coordination and electronic structure.</p>



<p class="wp-block-paragraph"><strong>6.4 Mass spectrometry</strong></p>



<p class="wp-block-paragraph">Mass spectrometry can assist in determining molecular mass and confirming the proposed molecular composition. High-resolution mass spectrometry is particularly valuable for distinguishing compounds with similar nominal masses.</p>



<p class="wp-block-paragraph"><strong>6.5 Elemental analysis</strong></p>



<p class="wp-block-paragraph">Carbon, hydrogen, nitrogen, sulfur, and metal content can be determined to assess whether the experimentally obtained composition agrees with the proposed molecular formula.</p>



<p class="wp-block-paragraph"><strong>6.6 X-ray crystallography</strong></p>



<p class="wp-block-paragraph">Single-crystal X-ray diffraction provides the most direct method for establishing molecular and coordination geometry when suitable crystals can be obtained. It can reveal metal–ligand bond lengths, bond angles, coordination number, and spatial arrangement of ligands.</p>



<p class="wp-block-paragraph"><strong>6.7 Thermal analysis</strong></p>



<p class="wp-block-paragraph">Thermogravimetric analysis and differential scanning calorimetry can provide information regarding thermal stability, coordinated or lattice water, decomposition behavior, and phase transitions.</p>



<p class="wp-block-paragraph"><strong>7. Major Metal Centers Investigated for Anticancer Applications</strong></p>



<p class="wp-block-paragraph">Platinum remains the most extensively validated metal in anticancer chemotherapy, primarily because platinum complexes can form strong interactions with DNA. However, the limitations of platinum drugs have encouraged exploration of other metals.</p>



<p class="wp-block-paragraph"><strong>Ruthenium complexes</strong> are particularly promising because of their diverse oxidation states, adaptable coordination chemistry, and potential for selective accumulation in tumor tissues. Their mechanisms may include DNA interaction, redox modulation, inhibition of proteins, and disruption of cellular signaling.</p>



<p class="wp-block-paragraph"><strong>Copper complexes</strong> can exploit the redox activity of copper and may promote reactive oxygen species generation. Because copper is an endogenous element, appropriately designed copper complexes may offer opportunities for developing biologically compatible metal-based agents.</p>



<p class="wp-block-paragraph"><strong>Cobalt and iron complexes</strong> can participate in redox processes and oxygen-related biological pathways. <strong>Zinc complexes</strong> are particularly interesting because zinc is an essential biological metal and can influence enzyme activity and cellular signaling [7]. Palladium complexes share some chemical characteristics with platinum compounds but often exhibit different ligand-exchange kinetics and biological behavior. Nickel complexes have also been investigated, although their biological and toxicological properties require careful assessment.</p>



<p class="wp-block-paragraph"><strong>8. Mechanisms of Anticancer Activity</strong></p>



<p class="wp-block-paragraph">The anticancer activity of heterocyclic and coordination compounds can arise through several complementary mechanisms.</p>



<p class="wp-block-paragraph"><strong>DNA interaction</strong></p>



<p class="wp-block-paragraph">Planar aromatic heterocycles can interact with DNA through intercalation, groove binding, or electrostatic interactions. Metal complexes may additionally form covalent or coordinate bonds with nucleobase donor atoms.</p>



<p class="wp-block-paragraph"><strong>Oxidative stress</strong></p>



<p class="wp-block-paragraph">Some metal complexes undergo redox reactions that increase intracellular reactive oxygen species. Excessive oxidative stress can damage DNA, proteins, lipids, and cellular organelles, ultimately promoting cancer-cell death.</p>



<p class="wp-block-paragraph"><strong>Apoptosis induction</strong></p>



<p class="wp-block-paragraph">Many heterocyclic and coordination compounds induce apoptosis by altering mitochondrial membrane potential, increasing pro-apoptotic signaling, activating caspases, or modifying the balance between pro- and anti-apoptotic proteins.</p>



<p class="wp-block-paragraph"><strong>Cell-cycle arrest</strong></p>



<p class="wp-block-paragraph">Bioactive compounds may interfere with regulatory proteins controlling cell-cycle progression, resulting in arrest at specific phases such as G₀/G₁, S, or G₂/M.</p>



<p class="wp-block-paragraph"><strong>Enzyme inhibition</strong></p>



<p class="wp-block-paragraph">Heterocyclic scaffolds can interact with kinase domains, topoisomerases, carbonic anhydrases, proteases, and other enzymes involved in cancer progression.</p>



<p class="wp-block-paragraph"><strong>Mitochondrial dysfunction</strong></p>



<p class="wp-block-paragraph">Some metal complexes accumulate within mitochondria and alter membrane potential, ATP production, or mitochondrial permeability, thereby initiating intrinsic apoptotic pathways.</p>



<p class="wp-block-paragraph"><strong>9. <em>In Vitro</em> Evaluation of Anticancer Activity</strong></p>



<p class="wp-block-paragraph">The initial biological evaluation of newly synthesized compounds is commonly performed using cancer cell lines representing different tissue origins. Examples include breast, lung, colon, liver, prostate, cervical, and hematological cancer models. The MTT assay is frequently used to estimate cellular metabolic activity following exposure to test compounds. Other assays, including resazurin-based methods, ATP-based viability assays, and clonogenic assays, can provide complementary information. The half-maximal inhibitory concentration (IC₅₀) is commonly calculated to compare the potency of compounds [8]. However, cytotoxicity against cancer cells should ideally be evaluated alongside normal or non-transformed cells to determine selectivity. Advanced studies may additionally investigate apoptosis using Annexin V/propidium iodide staining, caspase assays, mitochondrial membrane-potential measurements, and cell-cycle analysis by flow cytometry.</p>



<p class="wp-block-paragraph"><strong>10. Structure–Activity Relationships</strong></p>



<p class="wp-block-paragraph">Structure–activity relationship analysis is essential for identifying chemical features responsible for anticancer activity. Substituent type, position, electronic properties, steric characteristics, lipophilicity, heterocyclic ring structure, and metal identity can all influence biological performance. Electron-donating and electron-withdrawing substituents may alter ligand electronics and metal-binding properties. Halogen substitution can influence lipophilicity and cellular uptake, whereas hydroxyl and amino groups may improve hydrogen bonding and metal coordination. In coordination compounds, anticancer activity may depend not only on the ligand but also on metal oxidation state, coordination geometry, ligand-exchange kinetics, aqueous stability, and intracellular speciation. Consequently, the biologically active species may sometimes differ from the original compound administered in the experimental system.</p>



<p class="wp-block-paragraph"><strong>11. Role of Heterocyclic Ligands in Metal-Based Anticancer Agents</strong></p>



<p class="wp-block-paragraph">Heterocyclic ligands can significantly modify the pharmacological properties of metal complexes. A ligand can determine the stability of a complex, influence its solubility and lipophilicity, control ligand-exchange reactions, and facilitate interactions with specific biomolecular targets. Schiff bases containing nitrogen and oxygen donor atoms are particularly useful because they can form stable chelate complexes with several transition metals. Thiosemicarbazone derivatives are another important class because their sulfur and nitrogen donor atoms provide strong metal-binding capability while the organic framework itself can exhibit biological activity. The combination of a biologically active heterocycle with an appropriate metal center may therefore produce synergistic activity, although this assumption must be experimentally demonstrated rather than inferred solely from chemical structure.</p>



<p class="wp-block-paragraph"><strong>12. Selectivity and Toxicity Considerations</strong></p>



<p class="wp-block-paragraph">Although many heterocyclic and metal coordination compounds demonstrate promising anticancer activity in vitro, high cytotoxicity alone does not establish therapeutic potential. A successful anticancer agent should ideally exhibit preferential activity toward malignant cells while minimizing damage to normal tissues. Metal complexes require particular attention because metal release, ligand exchange, accumulation, and metabolism can influence toxicity. Studies should therefore evaluate hemolysis, normal-cell cytotoxicity, genotoxicity, oxidative stress, and relevant organ toxicity where appropriate.</p>



<p class="wp-block-paragraph">The therapeutic index and selectivity index can provide more meaningful information than cancer-cell IC₅₀ values alone. Comprehensive biological evaluation is therefore essential before advancing promising compounds toward animal or clinical studies.</p>



<p class="wp-block-paragraph"><strong>13. Recent Strategies for Improving Anticancer Performance</strong></p>



<p class="wp-block-paragraph">Several approaches are being investigated to improve the therapeutic potential of heterocyclic and coordination compounds. Ligand modification can improve solubility, cellular uptake, target recognition, and selectivity. Incorporation of targeting groups may promote preferential accumulation in tumor-associated environments. Nanotechnology-assisted delivery offers another strategy for improving the pharmacological performance of poorly soluble or unstable compounds. Metal complexes and heterocyclic agents can potentially be incorporated into polymeric nanoparticles, liposomes, dendrimers, or other nanocarriers. Stimuli-responsive delivery systems can release active compounds in response to tumor-associated characteristics such as acidic pH, elevated reactive oxygen species, or specific enzymes. Combination therapy involving metal complexes and established chemotherapeutic agents is also being investigated to overcome drug resistance and achieve complementary mechanisms of action.</p>



<p class="wp-block-paragraph"><strong>14. Challenges and Future Perspectives</strong></p>



<p class="wp-block-paragraph">Several challenges must be addressed before heterocyclic and coordination compounds can achieve broader clinical translation. Poor aqueous solubility, limited tumor selectivity, instability under physiological conditions, rapid metabolism, nonspecific toxicity, and inadequate pharmacokinetic properties can restrict therapeutic development. For metal complexes, understanding their behavior in biological fluids is particularly important because protein binding, ligand exchange, redox transformations, and metal release may substantially alter their biological activity. Future research should prioritize mechanism-based drug design, systematic structure–activity relationship studies, computational modeling, advanced pharmacokinetic evaluation, and appropriate <em>in vivo</em> validation. Greater emphasis should also be placed on evaluating selectivity toward cancer cells, rather than simply maximizing general cytotoxicity. The integration of medicinal chemistry with coordination chemistry, molecular biology, nanotechnology, and computational drug discovery may facilitate the development of next-generation compounds with improved efficacy and reduced toxicity.</p>



<p class="wp-block-paragraph"><strong>15. Conclusion</strong></p>



<p class="wp-block-paragraph">Heterocyclic compounds and metal coordination complexes represent highly versatile platforms for anticancer drug discovery. Heterocyclic scaffolds provide structural diversity and favorable molecular recognition characteristics, whereas metal coordination introduces additional opportunities for controlling redox activity, DNA interaction, enzyme inhibition, mitochondrial function, and cellular signaling. Advances in synthetic chemistry have enabled the development of increasingly diverse ligand frameworks and coordination architectures involving metals such as ruthenium, platinum, copper, cobalt, palladium, zinc, and nickel. Comprehensive characterization using spectroscopic, analytical, thermal, and crystallographic techniques is essential for establishing molecular structure and coordination behavior. Biological studies indicate that these compounds can induce apoptosis, oxidative stress, DNA damage, cell-cycle arrest, and inhibition of cancer-associated pathways. Nevertheless, promising in vitro cytotoxicity must be accompanied by evidence of selectivity, stability, pharmacokinetic suitability, and acceptable toxicity. Future development should therefore emphasize rational ligand design, mechanism-based screening, targeted delivery, and systematic in vivo validation. The convergence of heterocyclic medicinal chemistry and coordination chemistry is expected to remain an important strategy for discovering novel anticancer agents capable of addressing therapeutic resistance and limitations associated with existing treatments.</p>
